Recognizes a protein of 54 kDa, which is identified as cyclin A. Its epitope is located amino acids 144-148 of human Cyclin A2. Cyclins are regulatory subunits of the cyclin-dependent kinases (cdk's) and they control transition at different specific phases of the cell cycle. The temporal expression of cyclins is tightly regulated and subsequently plays a critical role in controlling the enzymatic activity of cdk's. These cyclin/cdk complexes are essential for passage through specific stages in the cell cycle. In mammalian somatic cells, cyclin A is required for S-phase and passage through G2-phase. The D and E type cyclins regulate the passage of G1, while cyclin B is a critical regulator of mitosis. Mutation or disruption of normal cyclin A expression causes cells to arrest in G2-phase. Carbonic anhydrase VIII (carbonic anhydrase 8) was named because of sequence similarity to other known carbonic anhydrase genes. However, the gene product lacks carbonic anhydrase activity (i. e. , the reversible hydration of carbon dioxide). The gene product continues to carry a carbonic anhydrase designation based on clear sequence identity to other members of the carbonic anhydrase gene family. Aldo-keto Reductase Family 1 Member B1 (AKR1B1), also known as aldose reductase or aldehyde reductase, is a member of the aldo/keto reductase superfamily, which consists of more than 40 known enzymes and proteins. This protein catalyzes the reduction of a number of aldehydes, including the aldehyde form of glucose, and is thereby implicated in the development of diabetic complications by catalyzing the reduction of glucose to sorbitol. It has also been shown to have decreased expression in adrenocortical cancer, and possibly play a role in adrenal tumorigenesis. CD40 is a receptor on antigen-presenting cells of the immune system and is essential for mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG-72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma. PAI-RBP1 (plasminogen activator inhibitor 1 RNA-binding protein), also known as SERBP1 (SERPINE1 mRNA-binding protein 1), CGI-55, CHD3IP (chromodomain helicase DNA binding protein 3 interacting protein), HABP4L or PAI-RBP1, is a membrane-associated protein that localizes to the nucleus, the perinuclear region of the cytoplasm and the plasma membrane. PAI-RBP1 is believed to play a role in the regulation of mRNA stability, as it specifically binds to the CRS (cyclic nucleotide-responsive sequence) motif of the PAI-1 mRNA and acts to stabilize the mRNA and regulate its expression. In addition, PAI-RBP1 interacts with Mi2- and may be involved in chromatin remodeling. PAI-RBP1 also interacts with PGRMC1 and participates in the transduction of Progesterone's antiapoptotic action in ovarian cell types. Recognizes a protein of 110 kDa, identified as Excision Repair Cross Complementing 1 (ERCC1). It is a mammalian nucleotide excision repair (NER) enzyme involved in repair of damaged DNA. ERCC1 is a homologous to RAD10 in Saccharomyces cerevisiae, which is required in mitotic intrachromosomal recombination and repair. EBAG9, also known as RCAS1, is an estrogen-transcribed protein. Soluble and membranous RCAS1 proteins may play a role in the immune escape of tumor cells by promoting T lymphocyte inhibition of growth and apoptosis. RCAS1 is expressed in a wide variety of cancers, including uterine, ovarian, and lung cancer cells, and acts as a ligand for a putative receptor present on peripheral lymphocytes. RCAS1 is highly expressed not only in cancer cells but also in non-tumor bile duct cells subject to immune attack. RCAS1 inhibits the in vitro growth of receptor-expressing cells and induces apoptosis, contributing to the ability of tumor cells to evade host immune surveillance. This MAb reacts with a 68 kDa protein, identified as light sub-unit of neurofilaments (NF-L). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.

This antibody recognizes a protein of 55-62 kDa, identified as cyclin B1. In mammals, cyclin B associates with inactive p34cdc2, which facilitates phosphorylation of p34cdc2 at aa 14Thr and 15Tyr. This maintains the inactive state until the end of G2-phase. The inactive cyclin B-p34cdc2 complex continues to accumulate in the cytoplasm until the completion of DNA synthesis, when Cdc25, a specific protein phosphatase, dephosphorylates aa 14Thr and 15Tyr of p34cdc2 rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when cyclin B is degraded. This degradation process is ubiquitin-dependent and is necessary for the cell to exit mitosis. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer.
